The Brazilians suffered a shock 2-1 loss to Chippa United in the Nedbank Cup last-32 last weekend.
A win against Usuthu tonight will see them claiming top spot in the league as they sit three points behind log leaders Bidvest Wits.
AmaZulu were also knocked out of the Nedbank Cup last weekend by Highlands Park on penalties.
They currently sit in 12th position in the standings with 17 points from 18 matches.
