The incident took place on Friday at an address in Tsakane Extension 15 at around 5am.
Tsakane police spokesman Capt Petros Mabuza says the suspect allegedly had an argument with his wife (52) and stabbed her to death.
Police were alerted to the incident by neighbours.
Mabuza says the woman was declared dead on the scene.
The suspect was arrested and is expected to appear in the Tsakane Magistrate Court soon.
